چکیده

Kidney and esophageal tumors induced by alkylating /V-nitroso com pounds in rats contain a high incidence (75-100%) of G -> A transition mutations in the p53 gene. These are almost selectively (89%) located in the tirsi base of codon 204 and the second base of 213, leading to amino acid substitutions Glu -»Lys and Arg -»Gin, respectively. In contrast to human neoplasms, a considerable fraction of rat kidney and esophageal tumors carries multiple p53 mutations. All nephroblastomas induced by transplacental exposure to /V-nitrosoethylurea and 56% of esophageal tumors induced by /V-nitrosomethylurea showed double mutations in codons 204 and 213 of exon 6. The selective targeting of p53 codons by alkylating nitrosamines may provide a basis for molecular epidemiolÃ3gi ca! studies on this class of chemical carcinogens.

New p53 Gene Mutation in non-Cancerous Mustard Gas Exposed Lung

Objective Mustard gas (MG) is a poisoning chemical, mutagenic and carcinogenic alkylating agent. It is used during World War I and also Iran-Iraq conflict. The p53 tumor suppressor gene is involved in the pathogenesis of malignant disease. The aim of this study is to determine possible mutation in p53 gene of lung sample from mustard gas exposed patients. Material and Methods Twelve lung bio...

Investigation of p53 and p27 expressions in the N-nitroso-N-methylureainduced breast cancer in female Wistar Albino rats

Introduction: N-nitroso-N-methylurea (NMU) is a carcinogen from nitrosamines family, which has been used to induce breast cancer in rodents. This model of breast cancer is very similar to the estrogen dependent breast cancer in human. As a continuation of our recent works, in the present study, the expressions of both p53 and p27 were investigated in NMU-induced breast cancer in Wistar Albin...

A rodent model for Wilms tumors: embryonal kidney neoplasms induced by N-nitroso-N'-methylurea.

Embryonal kidney cell tumors develop in rats given the alkylating agent N-nitroso-N'-methylurea as neonates. These tumors resemble the childhood Wilms tumors in their histopathology. Deletions and mutations in the Wilms tumor suppressor gene, WT1, are present in up to 6% of childhood nephroblastomas. To investigate the role of WT1 in rat kidney tumorigenesis, we studied the genetic alterations ...

شناسایی جهش در اگزون‌های پنج و شش ژن P53 در زنان مبتلا به سرطان پستان آذربایجان شرقی

Background and Objectives: Breast cancer (BC) is the most common invasive malignancy affecting women worldwide. The tumor-suppressor P53 gene (P53) is frequently mutated in breast tumors. To use P53 as a target for therapy, it is important to accurately assess p53 mutation status in tumor samples. Materials and Methods: A total of 102 tumor samples were collected from breast cancer patients ref...
